AlphaThera Product Spotlight: Antibody-DNA conjugates with oYo-Link™ Oligo

AlphaThera recently launched a powerful new bio-conjugation product enabling researchers to site-specifically label their antibodies with synthetic strands of DNA, otherwise known as oligonucleotides. The oYo-Link Oligo product can be used in advanced research applications including subcellular feature analysis with multiplexing, ultra high sensitivity protein detection with Immuno-PCR, and other next-gen single cell analysis techniques. The product launch has garnered interest from clients across the world.

How it Works:

oYo-Link Oligo enables precise attachment of either single-stranded or double-stranded DNA with user-defined sequences to the researchers’ antibody of choice. This is enabled by AlphaThera’s proprietary oYo-Link conjugation technology stemming from the publication titled “LASIC: Light Activated Site-Specific Conjugation of Native IgGs”, by Hui et.al. The DNA can be over 80 bases in length and are user-defined. The oYo-Link site-specific bioconjugation method (Fig 1) ensures that the customer’s oligonucleotide attaches precisely to the Fc-constant region of their antibody, guaranteeing well-defined antibody-oligonucleotide conjugate ratios and improved antibody binding performance. 

Applications of this product are continuing to garner enthusiasm in the research community. Antibody-oligo conjugates combine the specificity of antibodies with the amplification power of DNA sequencing to truly offer a next-generation assay tool. The antibodies bind specific target cell populations or subcellular features, and the attached oligonucleotide offers a method of identification and quantification of this bound target. Some of the applications of oYo-Link Oligo include advanced multiplexing, ultrasensitive Immuno-PCR, and Proximity Ligation Assays (Fig 2).

Multiplexing by Antibody Barcoding or Hashtagging: Antibody-oligonucleotide conjugates leverage the programmability of DNA barcodes to enable high-resolution cellular and subcellular feature analysis provided by multiplexing capabilities. Hundreds of subcellular targets can be effectively labeled and detected.

Immuno-Pcr (iPCR): A highly powerful immunodetection method that combines ELISA specificity with the signal amplification power of PCR. Enabled by the Antibody-DNA conjugate, iPCR detection assays have shown to be 100-1000 fold more sensitive than traditional ELISA with much less antibody used.

Proximity Ligation Assays: Utilize antibody-oligonucleotide conjugates to create a highly specific and sensitive immunohistochemical tool, enabled by a pair of DNA proximity probes conjugated to antibodies. This tool couples the specificity of ELISA with the powerful sensitivity of PCR to study single proteins, protein-protein interactions, post-translational modifications and more, for high-sensitivity detection of protein interactions.
